**Purpose:**
This ELISA kit is designed for the quantitative determination of Factor VIII-Related Antigen (FVIII-Ag) in human serum, plasma, and other biological liquid samples. It provides a reliable and efficient method for assessing FVIII-Ag levels, which are critical in diagnosing and monitoring coagulation disorders such as hemophilia A.
**Experimental Principle:**
The kit employs a double-antibody sandwich ELISA technique to detect FVIII-Ag in the sample. The microtiter plate is pre-coated with a specific monoclonal antibody against FVIII-Ag. After incubation with the sample, the FVIII-Ag present in the sample binds to the immobilized antibody. A horseradish peroxidase (HRP)-labeled secondary antibody is then added, forming a complex of antibody-antigen-enzyme-labeled antibody. Following thorough washing steps, the substrate TMB (3,3',5,5'-tetramethylbenzidine) is introduced. Under the catalytic action of HRP, TMB turns blue and then changes to yellow upon addition of an acidic stop solution. The intensity of the color developed is directly proportional to the concentration of FVIII-Ag in the sample. The optical density (OD) is measured at 450 nm using a microplate reader, and the FVIII-Ag concentration is determined by comparing the OD values to a standard curve.
