A new type of bacterial carbapenem antibiotic hydrolase was identified

--Recently, a research report "GES-18, a new carbapenem-hydrolyzing GES-type β-lactamase from Pseudomonas aeruginosa that contains Ile80 and Ser170 residues." Published in the recent international journal Antimicrobial Agents and Chemotherapy Researchers at Wenzhou University and others have revealed a new carbapenem antibiotic hydrolase GES-18 found in Pseudomonas aeruginosa.

The new antibiotic hydrolase was isolated from a clinical strain. The researchers isolated a clinical strain from an 81-year-old man with a lower respiratory tract infection from Belgium.

The researchers used PCR sequencing technology to analyze the strains and identified a new type of GES mutant, named GES-18, which is different from the carbapenem hydrolase GES-5 and GES-1. After detailed kinetic analysis, the investigator Kurt M. Hoffmann said that GES-18 and GES-5 can hydrolyze antibiotics imipenem and cefoxitin according to similar kinetic parameters. Compared with GES-1, GES-18 showed lower sensitivity to hydrolysis of β-lactamase inhibitors such as clavulanate and tazobactam.

The structure of GES-18 is similar to the hydrolyzed structure of GES-1 and GES-2, while the residues Val80Ile and Gly170Ser can trigger a more elaborate rearrangement of their structure. GES-1 analogs can be easily removed.

In conclusion, the researchers' kinetic analysis and crystallographic results revealed that GES-18 can be distinguished from GES-5 by substitution of Gly170Ser residues. This is very important for further research on bacterial resistance.
