Scope of application

Structure and function

Installation Precautions

1. The whole machine should be placed in a place where the temperature difference between the temperature is small and the operation is convenient. Avoid direct sunlight and keep away from the heating equipment. Place it in a stable manner.
2. Mix the gas cylinders and nitrogen cylinders smoothly and install relief valves (including pressure gauges). Place in place.
3. Connect the air path and check the air path. To prevent air leakage, seal the joints at each take-over place if necessary.

Operation room anaerobic environment formation

1) Place necessary accessories and utensils as required and put two nontoxic plastic bags into the operating room.
2) Adjust the input pressure of the mixed gas cylinder and nitrogen bottle and adjust the pressure reducing valve so that the input pressure is 0.1Mpa.
3) After the device is turned on, after the power switch, press the main power button on the control board to power on the device.
4) Place 1000 g of palladium particles (seal) into the chamber, dehumidify the condensing system, and place the anaerobic indicator.
5) Close sampling indoor and outdoor doors and vacuum check. (Press the vacuum pump key on the control panel, press it once, then press it again)
6) First replacement in the operating room (nitrogen replacement):
(1) Insert the rubber tube into the air inlet of the operating room and insert the plastic bag.
(2) Switch on the nitrogen inlet and open the nitrogen control valve in the operating room. After filling the two plastic bags with nitrogen, close the solenoid valve and tie the bag.
(3) Put the latex glove on the flange of the viewing plate and tighten it.
(4) Gradually put the nitrogen in the plastic bag in the operation room until it is completely controlled.
7) The operation chamber is replaced twice (nitrogen replacement) and the nitrogen flushing process is repeated. The sampling chamber is first evacuated and the exhaust valve is closed at any time.
8) Replacement of the operating room for the third time (mixed gas replacement) (The mixed gas ratio is H2 1/85% H2 1/10% H2 1/10% CO2/15%)
(1) Changing the gas path Open the mixing valve inlet of the operating room. When the gas is flushed, the sampling chamber is first evacuated, and at the same time, press the exhaust valve to close the exhaust.
(2) After the gas mixture fills the plastic bag, close the solenoid valve and open the mixture gas flow restriction valve so that the mixture gas passes through the flow stabilizer, the flow meter, and the flow meter. The flow rate is about 10 ml per minute.
(3) Gradually arrange the air-fuel mixture in the plastic bag in the operating room.
(4) After three times of ventilation, the oxygen content of the gas in the operating room is already in an extremely large state.
9) Open the palladium particle oxygen scavenger in the operation room, turn on the deoxygenation catalyst to deoxidize the catalyst, and dribble clean water on the anaerobic indicator strip reaction zone. After 1-2 hours, observe the discoloration and the reaction part. Dark blue turns light blue into an anaerobic environment in the operating room.
10) Ultraviolet sterilizing lamps are sterilized in the room and the sterilizing time is self-defined.
Inoculation and culture of strains

1) Check the sampling room door and close it.
2) Open the sampling door and close the bacteria after putting the bacteria in the sampling chamber.
3) The sampling chamber is filled with nitrogen and replaced by three processes. The vacuum is first stopped at a vacuum of 500 mmHg (66Kpa) and then the oxygen valve of the sampling chamber is opened to return the pointer to zero and the valve is closed. The sample chamber was filled with nitrogen and replaced three times.
4) If the selected vacuum is lower, the number of replacements needs to be increased.
5) After the sampling outdoor door is opened and closed, check the low vacuum 100 mmHg (13Kpa).
6) The conditions for long-term continuous use of anaerobic incubator (1) Put the Meilan indicator in the operating room every day. If it is not normal, it must be re-ventilated.
(2) To continuously input a small amount of mixed gas for a long period of time. The supplemental nitrogen can be combined with trace amounts of nitrogen through catalytic absorption to ensure an anaerobic state in the chamber. The flow rate of the supplemental mixed gas is selected to be about 10 ml per minute.
(3) Continuous culture runs for one day, replace the oxygen scavenger and desiccant. (Replaced oxygen scavenger and desiccant can be placed in 200 °C drying oven, drying it for 2-3 hours to restore it)
7) Inoculation rod sterilizer application:
(1) At any time, the electrode is short-circuited with a nickel heating wire to become red heat for sterilization (inoculation rod).
(2) Molten wax can be placed directly on the tube seal wax to rotate melted wax.
8) The temperature in the incubator of the operating chamber incubator can be arbitrarily selected and controlled.
Precautions

1. As far as possible, the instrument is installed in a place where the air is calm and where the temperature change is small.
2. Before starting the machine, you should fully familiarize yourself with and understand the specifications of the various supporting instruments and meters and master the correct methods of use.
3. The culture insert must be placed in the operating room after reaching an absolute anaerobic environment.
4. If there is a failure (for reasons such as stopping the gas), the anaerobic state can still be maintained for 12 hours in the operating room. (If more than 12 hours, remove the culture as needed for another treatment).
5. Always pay attention to air leakage.
6. When changing gas cylinders, pay attention to tighten the gas pipes and avoid inflow of oxygen-containing gas
7. Use the vacuum pump as required and check the fuel regularly.
8. Stop use, turn off the main power button, and the power switch on the rear of the device
